Comparision of Different Extracellular Matrix in Mcf7 Tumor Spheroid Formation

چکیده
Alginate hydrogels have already been extensively used in cell encapsulation, but few investigations have been done to compare cell proliferation and drug response in different extracellular matrices. We previously developed a novel microfluidics approach for tumor spheroid generation and testing: a cellladen core-shell bead suitable for several days of culture, which contains growing spheroids in varying concentrations of collagen, Matrigel, and alginate within a standardized volume. Here, we compared the growth of MCF7 cells in alginate, collagen-alginate, and Matrigel-collagen-alginate. Alginate gels containing both collagen type I and Matrigel were found to enhance MCF7 proliferation and tumor spheroid uniformity in contrast to alginate alone or collagen-alginate. We used second harmonic generation imaging to visualize the collagen distribution inside the cell-laden beads, and examined dose response in hypoxic conditions.

Comparison of radiosensitizing effect of Resveratrol on monolayer and spheroid culture of DU145 prostatic cell line
Background: Radiotherapy is an established therapeutic modality for prostate cancer. Resveratrol, a natural antioxidant, has been shown to inhibit carcinogenesis and to block the process of tumor initiation and progression. No data is available on the response of cellular spheroid to Reseveratol. In this study we have examined the effect of Resveratol on the radiation response of human p...
متن کاملModelling Tumor-induced Angiogenesis: Combination of Stochastic Sprout Spacing and Sprout Progression
Background: Angiogenesis initiated by cancerous cells is the process by which new blood vessels are formed to enhance oxygenation and growth of tumor. Objective: In this paper, we present a new multiscale mathematical model for the formation of a vascular network in tumor angiogenesis process. Methods: Our model couples an improved sprout spacing model as a stochastic mathematical model of spro...
متن کاملParticipation of tenascin and transforming growth factor-beta in reciprocal epithelial-mesenchymal interactions of MCF7 cells and fibroblasts.
The tumor stroma is essential for the development of the tumor epithelium. Tenascin is an extracellular matrix protein highly expressed in the stroma of malignant mammary tumors. We therefore tested whether in vitro MCF7 cells were able to induce fibroblasts to synthesize tenascin. Indeed MCF7 cell-conditioned medium contained tenascin-inducing activity. This activity was shown to be transformi...
متن کاملEmpirical chemosensitivity testing in a spheroid model of ovarian cancer using a microfluidics-based multiplex platform.
The use of biomarkers to infer drug response in patients is being actively pursued, yet significant challenges with this approach, including the complicated interconnection of pathways, have limited its application. Direct empirical testing of tumor sensitivity would arguably provide a more reliable predictive value, although it has garnered little attention largely due to the technical difficu...
متن کاملTwo Dimensional Mathematical Model of Tumor Angiogenesis: Coupling of Avascular Growth and Vascularization
Introduction As a tumor grows, the demand for oxygen and nutrients increases and it grows further if acquires the ability to induce angiogenesis. In this study, we aimed to present a two-dimensional continuous mathematical model for avascular tumor growth, coupled with a discrete model of angiogenesis. Materials and Methods In the avascular growth model, tumor is considered as a single mass, wh...
